Hepatitis C-like viruses identified in bats and rodents

Apr. 22, 2013 ? As many as one in 50 people around the world is infected with some type of hepacivirus or pegivirus, including up to 200 million with hepatitis C virus (HCV), a leading cause of liver failure and liver cancer. There has been speculation that these agents arose in wildlife and jumped species to infect humans; however, little was known about their distribution in other species.

In two new papers published in the journals mBio and Proceedings of the National Academy of Sciences, investigators at the Center for Infection and Immunity (CII) at Columbia University's Mailman School of Public Health report the discovery of hepaciviruses and pegiviruses -- close relatives of HCV -- in rodents and bats. The viruses are similar to those that infect humans and may therefore provide insights into the origins of HCV, as well as the mechanisms behind animal-to-human transmission. It may also enable development of new animal systems with which to model HCV pathogenesis, vaccine design, and treatment.

Both discoveries were made using high-throughput sequencing and other molecular methods for pathogen discovery pioneered at CII. Both represented multicenter global efforts.

As reported in mBio, Amit Kapoor, PhD, and colleagues screened more than 400 wild-caught rodents. Molecular analysis revealed the presence of hepaciviruses and pegiviruses closely related to those found in humans. "Importantly, the rodent hepaviviruses contained sequences that are thought to play a role in liver infection in HCV," says Dr. Kapoor, lead author of the study and assistant professor of Pathology and Cell Biology at Columbia University Medical Center. "We also found instances of a single animal infected with multiple hepaciviruses."

Such co-infections have also been observed with HCV in humans, suggesting that the immune response to HCV is different than with most viral infections -- a finding that has implications for vaccine design. "It also supports the potential use of rodent hepaciviruses in developing models for human disease," says W. Ian Lipkin, MD, John Snow Professor of Epidemiology and director of the CII.

Researchers from Rockefeller University, University of Edinburgh, University of Copenhagen, University of New Mexico, North Carolina College of Veterinary Medicine, Pennsylvania State University and the National Institutes of Health contributed to the study. Results appear online in mBio.

In a second study led by P. Lan Quan, PhD, molecular assays of 1,615 bats collected worldwide led to the identification of 83 novel hepaciviruses and pegiviruses, representing an infection rate of nearly 5%. "The broad prevalence, unprecedented diversity, and worldwide distribution of these novel viruses suggest that bats are a major and ancient reservoir for both hepaciviruses and pegiviruses, and provide insights into the evolutionary history of HCV and human pegiviruses," says Dr. Quan, associate research scientist at the Center for Infection and Immunity.

European pension proposals could cost UK business 450 billion pounds

LONDON (Reuters) - British businesses could face a 450 billion pound bill as a result of plans to force pension funds to protect themselves from risk with extra capital, data from Europe's insurance and pensions watchdog showed.

This could hamper companies' ability to invest and also force some pension schemes to close.

The estimate from the European Insurance and Occupational Pensions Authority (EIOPA) shows the impact of new capital requirements on final salary-linked pension schemes, where benefits are based on salary and duration of employment.

The 450 billion pound estimate is in line with a worst-case scenario contained in figures the UK Pensions Regulator produced for the British government last year.

"This confirms that any such new rules would harm businesses' ability to invest, grow and create jobs, and many more (pension) schemes could be forced to close," Minister of State for Pensions Steve Webb said in a statement on Tuesday.

The British pensions industry was also critical.

The National Association of Pension Funds said the high price tag would have a highly damaging effect for the retirement prospects of millions of UK workers.

"Businesses trying to run final salary pensions could be faced with bigger pensions bills to plug an astonishing 450 billion pound funding gap," Joanne Segars, chief executive at the NAPF, said.

The new capital rules, known as Solvency II, were originally aimed at the insurance industry but the European regulator is proposing to adapt them for the pensions sector to improve standards of governance, risk management, valuation and calculation of minimum capital requirements.

Britain had the highest funding requirement out of the countries covered in the European watchdog's assessment, which were Belgium, Germany, Denmark, Ireland, the Netherlands, Norway and Sweden.

Earlier on Tuesday, the European watchdog rejected calls for it to cut capital charges on insurance companies' infrastructure and private equity investments, which politicians are pushing as a way to boost long-term growth.

First-in-humans study introduces next generation cell therapy

ROCHESTER, Minn. -- Translating a Mayo Clinic stem-cell discovery, an international team has demonstrated that therapy with cardiopoietic (cardiogenically-instructed) or "smart" stem cells can improve heart health for people suffering from heart failure. This is the first application in patients of lineage-guided stem cells for targeted regeneration of a failing organ, paving the way to development of next generation regenerative medicine solutions. Results of the clinical trial appear online of the Journal of the American College of Cardiology.

The multi-center, randomized Cardiopoietic stem cell therapy in heart failure (C-CURE) trial involved heart failure patients from Belgium, Switzerland and Serbia. Patients in the control group received standard care for heart failure in accordance with established guidelines. Patients in the cell therapy arm received, in addition to standard care, cardiopoietic stem cells -- a first-in-class biotherapeutic. In this process, bone marrow was harvested from the top of the patient's hip, and isolated stem cells were treated with a protein cocktail to replicate natural cues of heart development. Derived cardiopoietic stem cells were then injected into the patient's heart.

"The cells underwent an innovative treatment to optimize their repair capacity," says Andre Terzic, M.D., Ph.D., study senior author and director of the Mayo Clinic Center for Regenerative Medicine. "This study helps us move beyond the science fiction notion of stem cell research, providing clinical evidence for a new approach in cardiovascular regenerative medicine."

Every patient in the stem cell treatment group improved. Heart pumping function improved in each patient within six months following cardiopoietic stem cell treatment. In addition, patients experienced improved fitness and were able to walk longer distances than before stem cell therapy. "The benefit to patients who received cardiopoietic stem cell therapy was significant," Dr. Terzic says.

In an accompanying editorial, Charles Murry, M.D., Ph.D., and colleagues at the University of Washington, Seattle, say, "Six months after treatment, the cell therapy group had a 7 percent absolute improvement in EF (ejection fraction) over baseline, versus a non-significant change in the control group. This improvement in EF is dramatic, particularly given the duration between the ischemic injury and cell therapy. It compares favorably with our most potent therapies in heart failure."

The science supporting this trial is a product of a decade-long journey in decoding principles of stem cell-based heart repair. "Discovery of rare stem cells that could inherently promote heart regeneration provided a critical clue. In following this natural blueprint, we further developed the know-how needed to convert patient-derived stem cells into cells that can reliably repair a failing heart," says Dr. Terzic, underscoring the team effort in this endeavor.

Initial discovery led to the identification of hundreds of proteins involved in cardiogenesis, or the heart development process. The research team then identified which proteins are necessary in helping a stem cell become a reparative cell type, leading to development of a protein cocktail-based procedure that orients stem cells for heart repair. Such upgraded stem cells are called cardiopoietic or heart creative.

Reducing waste of food: A key element in feeding billions more people

Apr. 7, 2013 ? Families can be key players in a revolution needed to feed the world, and could save money by helping to cut food losses now occurring from field to fork to trash bin, an expert said in New Orleans on April 7. He described that often-invisible waste in food -- 4 out of every 10 pounds produced in the United States alone -- and the challenges of feeding a global population of 9 billion in a keynote talk at the 245th National Meeting & Exposition of the American Chemical Society, the world's largest scientific society.

"We will need another 'Green Revolution' to feed the world by 2050," said John Floros, Ph.D., referring to the development of high-yield, disease-resistant breeds of grain and other agricultural innovations that took root in the 1960s. "That will mean scientific innovations, such as new strains of the big three grains -- rice, wheat and corn -- adapted for a changing climate and other conditions. It also will require action to reduce a terrible waste of food that gets too little attention."

Floros cited estimates that in many developing countries up to half of the food harvested from farmers' fields is lost before reaching consumers. He is dean of the College of Agriculture at Kansas State University. That waste can occur due to spoilage from improper storage of grain during transportation or from pests. Rats and mice alone eat or spoil 20 percent of the world's food supply due to contamination with their urine and feces.

"A different kind of waste occurs in the United States and some other developed countries," Floros said. "Developed countries have much more efficient systems for preserving, storing, transporting and protecting food from spoilage and pests. But as a nation -- households, supermarkets, restaurants, other food-service providers -- we throw away about 4 out of every 10 pounds of food produced each year."

Government studies show, for instance, that the average family in the United States throws away 20 pounds of food a month, more than $2,000 worth every year for a family of four. It includes food that has gone uneaten and spoiled in refrigerators and on pantry shelves, as well as food that people throw away after cooking. Uneaten food actually rivals paper, plastic and other refuse as the No. 1 material in some municipal landfills.

Scientists know that food waste in landfills, for instance, releases methane gas as it decomposes. Methane is about 20 times more potent than carbon dioxide as a greenhouse gas that fosters global warming. Floros pointed out that reducing food waste would contribute to solving other great global challenges that society faces in the 21st century, beyond feeding a booming population. Wasting food wastes the freshwater needed to grow it, at a time when 1.2 billion people lack access to clean water. It also wastes energy, fertilizers, pesticides and other resources used in the food supply.

Supplying more food, however, is only part of the challenge, Floros emphasized. "Millions of people in some developing countries are becoming more affluent. In the past, people were satisfied with food that filled them up and sustained life. Increasingly, they will demand food that is convenient to prepare, certified as safe and highly nutritious and tastes good."

He cited the People's Republic of China as an example. The middle class in China is now larger than the U.S. population and is increasing in size year by year. And people in China are now consuming almost 3 times as much meat compared to a few decades ago. Demand for convenience foods also is rising with the growth of the urban population.

Several other food-related challenges lie ahead, Floros pointed out. Water, for instance, is becoming scarcer, as is fertile farmland. Global climate change may stress those resources even further. The demand for sustainable energy may divert more cropland to production of crops for biofuel production. Economic conditions threaten less investment in agricultural research and development. Drought and other extreme weather could impact food production. And consumption of too much food and less nutritious foods underpins epidemics in obesity and type 2 diabetes.

"We're not doing enough to resolve these complex issues that are critical for providing 9-10 billion people with a nutritious diet," said Floros. "Consumers, industry, universities and governments all need to pitch in. The first step is more awareness of these issues and the need for action on multiple levels of society."

North Korea has moved a medium-range missile, which could hit targets in South Korea and Japan, to its east coast, a report says. The South Korean Yonhap news agency said on Thursday that Pyongyang's move was detected by the US and South Korean intelligence.

South Korean Defense Minister Kim Kwan-jin said the missile could reach a "considerable distance" but that it was not capable of reaching the US mainland. "It could be aimed at test-firing or military drills."

The missile is reportedly a Musudan and is believed to have an intended range of about 3,000 kilometers. Reports said the missile faced Japan.

Intelligence sources said North Korea might fire the missile around the middle of April, when people celebrate the birthday of Kim Il-sung, the founder of North Korea, according to Yonhap.

Japan says it will respond to any North Korean attack immediately and it is bracing for a "worst-case scenario."

Luigi's Mansion: Dark Moon (Nintendo 3DS)


I never played the first Luigi's Mansion, so I didn't know what to expect beyond the premise of the sequel, Luigi's Mansion: Dark Moon. In this $39.99 (direct)?Nintendo 3DS game, you play Mario's downtrodden brother who must vacuum up ghosts in abandoned haunted houses at the behest of a baby-headed professor. I'm fairly certain that description is accurate and not the result of drinking too much Mountain Dew and staring at a glasses-free 3D screen for hours. Regardless of what I was expecting, Dark Moon is an entertaining, light adventure game that gives Luigi something to do even if that something isn't anywhere close to crawling out from under his brother's shadow.

Many Mansion
Instead of one mansion, Luigi has to explore several to get rid of the ghosts that have become aggressive because the crystal Dark Moon was mysteriously destroyed. This means playing through several stages that take place in different parts of each mansionn (with generous overlap and some backtracking) solving puzzles, then culminating in a boss fight to get a piece of the Dark Moon and clean out the mansion completely. The regular stages involve exploring increasingly large swaths of the mansion, unlocking new areas by getting new items or solving certain puzzles, like turning on generators.

Luigi doesn't have the jumping moves of Mario, but thanks to the Poltergust 5000 he has plenty of tools at his disposal. He can use the vacuum to suck in ghosts or pull away obstacles, and attached bulbs can stun ghosts, activate switches, and make invisible things appear. These tools factor more into solving puzzles than fighting ghosts, with each room full of sheets that can be sucked away, invisible objects that can appear, and doors to open.

In one area, I had to start a generator by flashing my light on four switches at the same time, but two would only appear if I was standing on one panel, and another two would only appear if I was standing on another. I used my vacuum to pick up a bucket and fill it with slime, then set it on the panel so I could make all switches appear. In another area, I had to use elastic goop that got stuck in my vacuum to work like a balloon to let me float to the next floor.

Ghost Busting
Besides solving puzzles, Luigi fights ghosts. The combat isn't quite as fun as the puzzle-solving, mostly focusing on getting past ghosts' defenses by waiting for them to strike or sucking away the different objects they use to protect themselves, stunning them, then wrestling them into the vacuum by pulling back and using charged strikes to weaken them enough to finally capture them. This applies to nearly every enemy, and while they look and behave slightly differently (green ghosts use junk to protect themselves from the light, big red ghosts come right at you with fists, blue ghosts hide in objects to avoid you) the actual ghost-hunting all feels very similar.

The exploration is important for finding money as well as advancing in the game. You collect coins, dollars, and gold bars hidden through each mansion, and can use them to get upgrades for the vacuum, which which appear automatically when you collect a certain amount of money. They can make the vacuum more powerful when fighting ghosts or otherwise more helpful without being vital to the game (the actual parts that solve puzzles are unlocked by playing the game itself). Money can be hidden anywhere, and only by interacting with, flashing, and shining the invisibility detector light on everything can you find all of it. Large caches are money are hidden particularly carefully, like behind walls that can only be accessed by sucking away loose wallpaper to expose holes, or by searching for keys to locked areas and taking detours around your main goal.

Multiplayer Modes
Dark Moon offers a surprisingly fun multiplayer aspect in the Scarescraper, a randomly generated series of floors one to four Luigis can tackle over local or online multiplayer. Each floor has a series of rooms with different objects to interact with, ghosts to find, and obstacles to get past, and depending on the mode you're playing you might have to hunt every ghost on the level, run through it as fast as possible, or find invisible ghost puppies. It doesn't have the big, crafted set piece puzzles like in the main quest, but the random element adds a layer of replayability to the game and makes multiplayer engaging.

Luigi's Mansion: Dark Moon is an entertaining adventure game and a long-awaited sequel to a game few even thought would see a sequel. It isn't very deep, but the puzzles, action, and multiplayer modes make it feel like a worthwhile package for any 3DS owner looking for something slightly different.?

Discovery Slate Includes Live High-Wire Crossing of Grand Canyon

NEW YORK (TheWrap.com) - Discovery's upcoming shows include a live special in which self-described "King of the High Wire" Nik Wallenda will cross the Grand Canyon without a harness.

"Wallenda Live" is one of the highlights (no pun intended) of a slate that also includes the previously announced scripted miniseries "Klondike" and another live special, "Fast N' Live," in which competitors will race to rebuild cars, albeit not while suspended 1,500 feet over the Little Colorado River. Oh well.

Discovery also announced four natural history specials or series, including the return of "Shark Week," and seven new series. Two of them are about searching for oil, and three focus on survivalists.

The Grand Canyon tightrope walk follows Wallenda's successful crossing of Niagara Falls last year.

Discovery is unveiling its new slate to advertisers as it boasts that it is among the top 10 cable networks in total primetime viewers.

"I am excited by our Upfront programming slate that includes our first ever scripted mini-series and bold live specials", said Eileen O'Neill, group president of Discovery and TLC Networks. "The past year has been filled with provocative and compelling characters, landmark live programming and storytelling ranging from epic to everyday. The best is yet to come."

SCRIPTED MINI-SERIES

KLONDIKE - Discovery Channel's first ever scripted mini-series, based on Charlotte Gray's book Gold Diggers: Striking It Rich in the Klondike. KLONDIKE brings to life the tale about six strangers and their collective fight for survival and wealth in a small, frontier town in the remote Klondike. KLONDIKE is a co-production between Discovery, Entertainment One Television (eOne) and Nomadic Pictures in association with Scott Free Television.

LIVE SPECIALS

MTV: Crew wasn't with 'BUCKWILD' star when he died

SISSONVILLE, W.Va. (AP) ? Production crews were not with a young star of the reality show "BUCKWILD" when he died over Easter weekend and hadn't filmed him since earlier in the week, MTV officials said Wednesday.

Shain Gandee, 21, was found dead Monday in his SUV near Sissonville, along with two passengers.

Autopsies determined that all three died of carbon monoxide poisoning, possibly caused by the vehicle's tailpipe being submerged in mud. That could have allowed the invisible gas to fill the vehicle's cabin.

MTV spokesman Jake Urbanski said it would be at least a few weeks before producers and cast members decide whether to continue shooting season two. For now, he said, everyone is focused on supporting Gandee's friends and family.

Also killed were his 48-year-old uncle, David Gandee, and 27-year-old Donald Robert Myers.

Cunningham-Parker-Johnson Funeral Home in Charleston said services for Shain and David Gandee were scheduled for 2 p.m. Sunday at the Charleston Municipal Auditorium. A private family burial service will follow in Thaxton Cemetery.

Family members said David Gandee worked for Appalachian Tire Co., was a fan of both the Pittsburgh Steelers and the West Virginia University Mountaineers, and loved golf.

Shain Gandee's family has requested that instead of flowers, fans and friends send donations to one of two charities: the Angels Among Us program at Duke University Medical Center in Durham, N.C., or the Autism Society in Bethesda, Md.

Track who's supporting same-sex marriage now

The cascade of senators announcing or clarifying their support for same-sex marriage in the past two weeks has somewhat obscured the fact that more than half of the Democrats in the Senate backed the policy before it became fashionable. By the time Vice President Joe Biden abruptly announced his own support for the policy on May 5, 2012, nearly two-dozen senators had already signed on to a campaign begun several months earlier calling for marriage equality to be included on the Democratic Party's platform. Another eight, including independent Bernie Sanders of Vermont, expressed their support prior to 2012.

The following chart tracks each Democratic senator's position on same-sex marriage organized around when he or she made the announcement. In some cases, this date is ambiguous. After the Huffington Post listed Michigan Democrat Debbie Stabenow among those who had not formally supported the measure, for example, her office indicated that she had already done so as part of her 2012 reelection campaign. Wikipedia has a highly comprehensive page on the subject that served as a basis for many of the links in this chart, all independently verified.

The chart also includes notable Democratic leaders outside the Senate as well as Republican and independent senators who have endorsed same-sex marriage.

SPOKANE, Wash. (AP) ? California coach Lindsay Gottlieb was in elementary school the last time a West Coast women's basketball program not named Stanford reached the Final Four.

Gottlieb understood the drought and the difficulty in getting to the national semifinals. That's partly why the grin on her face was so wide Monday night when Cal finally snapped that 25-year streak, sending the Golden Bears to New Orleans for the Final Four.

"These kids are brought in," Gottlieb said. "So many things go into it and then you have to get a little lucky and then things have to go right, so I'm really conscious of this is special."

Layshia Clarendon scored 17 of her 25 points in the second half and overtime, and California rallied from 10 down with less than 7 minutes left to beat Georgia 65-62 in the Spokane Regional final, advancing to the national semifinals for the first time in school history.

Long Beach State in 1988 was the last team from the western part of the country other than Stanford to reach the Final Four. During that 25-year span, eight different programs in the West reached the regional finals. But whether it was Long Beach State, Washington, USC, UCLA, Colorado, Utah, Arizona State or Gonzaga, they all came up one game short of advancing ? sometimes at the hands of Stanford.

Not these Golden Bears (32-3), who were picked by President Barack Obama to win the regional in his bracket.

"This is still better than my wildest dreams," Gottlieb said.

The loss ended Georgia coach Andy Landers' hope of reaching the Final Four for the first time since 1999. Georgia last reached the regional finals in 2004 and this was the Lady Bulldogs' 11th trip to the regional finals.

And it nearly became Landers' sixth Final Four.

"They made more plays than we did and they deserve to win the game," Landers said. "I'm disappointed, but I'm proud of our basketball team and the run that we have made."

Shacobia Barbee led Georgia (28-7) with 14 points and 10 rebounds, but the Lady Bulldogs' surprising postseason run that included an upset of Stanford came to a painful end.

"I don't think the words can even explain right now. Disappointed, hurt," said Georgia's Jasmine James, who had 11 points before fouling out. "To have something that you've always wanted to be able to do, just be so close, and to end up getting outworked for it, it hurts."

California will go to the Final Four on the side of the bracket completely blown open by the stunning loss of top-ranked Baylor in the regional semifinals. The Golden Bears will play Tennessee or Louisville in the national semifinals.

It didn't look like another game awaited the Golden Bears, who trailed 49-39 with 6:46 left after Barbee hit a pair of free throws. The Bears got back in it by halftime, overcoming a horrible shooting start, but each run early in the second half was rebuffed by the Lady Bulldogs. Cal missed 18 of its first 19 shots to start the game.

Finally Cal found an answer. Afure Jemerigbe hit her first 3-pointer after missing her previous six attempts. Clarendon scored four straight points and Talia Caldwell got a bucket in the paint. Caldwell then hit a putback of Jemerigbe's missed free throw to tie it at 50.

Tiaria Griffin missed from 3-point range for Georgia and Cal's Brittany Boyd was fouled driving to the rim by James, her fifth foul, leaving Georgia without its floor leader. Boyd hit both free throws and Cal had its first lead since 17-16.

Not having James on the floor had an immediate effect. Georgia turned it over on its next two possessions, helped by a pair of blocked shots by Jemerigbe, the second with 51 seconds left.

But the Bears' free throw struggles that nearly cost them in the second round against South Florida returned when Jemerigbe missed the front end of a 1-and-1 with 27 seconds left. Griffin missed another 3-point attempt, but Anne Marie Armstrong got the rebound and scored with 8.5 seconds left in regulation to force overtime. Georgia went up 55-52 when Khaalidah Miller hit a 3 to start overtime, but the Bears then took command. Jemerigbe hit a 3-pointer with 2:48 left to give Cal a 59-55 lead and Clarendon added a rebound putback for a 61-55 advantage with 1:26 remaining.

Barbee hit a pair of free throws with 1:03 left, but instead of fouling, Georgia chose to play defense. Clarendon made them pay, spinning away from Barbee and hitting a 15-footer with 37.9 seconds left for a 63-57 lead.

Georgia got within 64-62, but Clarendon split free throws with 2.1 seconds left and Barbee's desperation shot was off.

"I was smiling a lot in the second half even though we were losing," Clarendon said. "This is why you play basketball for these big moments. Really enjoy them and just relish them."

Burglar on crutches wanted for string of robberies

Police in Oklahoma City are searching for a man they say burglarized three homes?at least one of them while on crutches.

According to Oklahoma City's News 9, police say 28-year-old Ernest Sarabia hobbled up to the door of one home and pried it open. Once inside, he stuffed jewelry and electronics into a backpack before being chased away by the homeowner?sans crutches.

"When he leaves, he's seen being chased by the homeowner, who basically catches him in the act, chases him out," Sgt. Jennifer Wardlow of the Oklahoma City Police Department told the station. "He's hopping on one leg."

The suspect then fled the scene in a waiting truck, the video showed.

Police are asking anyone with information related to the hopping burglar to call 405-235-7300 or 1-800-632-TIPS.

There were 9,853 reported burglaries in Oklahoma City in 2012, according to crime statistics posted on the city's website. That number was virtually unchanged from 2011, when there were 9,863.

It's unclear how many of them were attempted while handicapped.
